Development of small molecule enhancers of autophagy for the clearance of protein aggregates and treatment of FTD

Frontotemporal dementia and other neurodegenerative disorders are characterized by the accumulation of protein in the brain cells. Although there are no known curative treatments for this disease, the removal of these proteins by a process called autophagy has been shown to be successful in a previously funded AFTD grant. Autophagy is a process where the cell activates its recycling system to get rid of redundant and potentially toxic proteins that build up over time. This application proposes to develop drugs that will activate this system in the brain in the hopes that this will remove the buildup of bad proteins in the brain cells and restore the individual to full function of the brain.
